چکیده

in which ground glass connections could not be used because of the large size of the flasks, it was found that benzene extracts of rubber stoppers previously washed with ether and methanol still contained blue-green fluorescing substances. These extracts had the ultraviolet absorption spectra of carcinogenic hydrocarbons and other compounds of interest. This discovery seemed of such great possible importance to investigators in cancer re search, to the rubber industry, and to public health that further experiments were made. It was found that extracts of rubber stoppers were car cinogenic for mice. In similar extracts, the pres ence of pyrene, 3,4-benzpyrene, chrysene, and other aromatic hydrocarbons was detected. The same compounds were found in benzene extracts of an automobile tire. Further observations, together with comments on the possible origins and impor tance of these compounds, are here presented.

International regulatory needs for development of an IATA for non-genotoxic carcinogenic chemical substances.

Although regulatory requirements for carcinogenicity testing of chemicals vary according to product sector and regulatory jurisdiction, the standard approach starts with a battery of genotoxicity tests. If any of the in vivo genotoxicity tests are positive, a lifetime rodent cancer bioassay may be requested, which allows the detection of non-genotoxic carcinogens (NGTxC). However, under most ch...

Implications of the lack of accuracy of the lifetime rodent bioassay for predicting human carcinogenicity.

The NTP lifetime rodent bioassay (LRB) is the "gold standard" for predicting human carcinogenicity. Unfortunately, little attempt has been made to validate it against human carcinogenicity. Here we show that the extremely limited data available do not support either of the two common interpretations of LRB results. If a risk-avoidance interpretation is used where any positive result in a sex/sp...

An overview of phthalate ester carcinogenicity testing results: the past.

This paper presents a short review of the available carcinogenicity tests which have been carried out on various phthalate esters prior to the availability of the National Cancer Institute bioassay studies. All of these studies suffer from diverse limitations in design or reporting when compared to more recent experimental protocols and reports. There are three papers related to di(2-ethylhexyl...

Validation of transgenic mice carrying the human prototype c-Ha-ras gene as a bioassay model for rapid carcinogenicity testing.

Carcinogenicity testing is indispensable for identifying environmental carcinogens and for evaluating the safety of drugs in the process of development. Conventional 2-year rodent bioassays are one of the most resource-consuming tests in terms of animals, time, and costs. Development of rapid carcinogenicity testing systems that can assess carcinogenicity within a short period has become a soci...

Regulatory requirements and ICH guidelines on carcinogenicity testing of pharmaceuticals: A review on current status

Carcinogenicity studies are conducted in laboratory animals to evaluate the carcinogenic potential of pharmaceuticals. In drug discovery, rodent carcinogenicity studies are conducted with an objective to explore the mechanism of carcinogenesis and the ultimate risk to humans. Many problems are encountered in carrying out and interpretation of results for carcinogenicity bioassay. A number of fa...

A Novel Strategy to Predict Carcinogenicity of Antiparasitics Based on a Combination of DNA Lesions and Bacterial Mutagenicity Tests

Genotoxicity and carcinogenicity testing of pharmaceuticals prior to commercialization is requested by regulatory agencies. The bacterial mutagenicity test was considered having the highest accuracy of carcinogenic prediction. However, some evidences suggest that it always results in false-positive responses when the bacterial mutagenicity test is used to predict carcinogenicity. Along with maj...
